Produção e caracterização de enzimas proteolíticas do cogumelo Ostra-Rei por fermentação submersa

As proteases são biocatalisadores com funções fisiológicas complexas, presentes em todos os seres vivos que predominam em processos biotecnológicos nos diversos segmentos da indústria. Das fontes naturais de protease, espécies de Pleurotus sintetizam compostos bioativos de interesse industrial, inclusive enzimas proteolíticas. Neste estudo foi investigada a produção e caracterização parcial de proteases de P. eryngii DPUA 1816 para futura aplicação industrial. A fermentação foi realizada em extrato de Malte ou Sabouraud + extrato de levedura 0,5% (p/v), por sete dias, a 28 ºC, 150 rpm. Nos extratos recuperados foi determinada atividade qualitativa de proteases e avaliado o efeito do pH, temperatura, íons e inibidores na atividade e estabilidade enzimática. Os resultados mostraram que, nas condições experimentais a atividade de proteases de P. eryngii foi 22,70 U/mL, com atividade ótima em pH 5,0, a 40 ºC, com estabilidade na faixa de 40 °C a 80 °C. Os íons Mn2+, Zn2+, Mg2+ e K+ aumentaram a atividade das enzimas em 85%, 84%, 40%, e 31%, respectivamente. Porém, Fe2+ e Cu2+ reduziram ação catalítica em 38% e 37%, respectivamente. O EDTA e o PMSF causaram inibição de 96% e 95,10%, respectivamente. Com base nesses resultados foi observada a presença de metaloproteases e serinoproteases, enzimas que tem importância industrial para elaboração de laticínios e fármacos.
